      Plasmid dna (pUC19) imaged in buffer with a liquid scanner (not a trivial task)       Rolling circle amplification (rca) product made at MU. pUC19 is about 3000 base pairs long, the rca product is single stranded dna up to 10kb in length.    
                   
 
                   

 

 

 
 
                   
      Rca product that has been incubated with biotinylated oligonucleotides that complement the rca sequence at approx. 30nm periodicity (one oligo hybridizes each 95 bases). The double stranded regions on the dna are clear as small swellings, the large (5nm) beads are streptavidin coated particles that do not seem to be well associated with the dna.
